The Y chromosome has areas that don't code for any genes (and so no health or disease information can be revealed by testing there). Some locations in the non-code areas have fast enough mutation rates to be useful in genealogy. The fast-mutating locations are repeated short sequences of DNA nucleotides. Some spots, called markers, have, for example, 23 repeats. This type of marker is called a Short Tandem Repeat or STR. Fast-mutating STR markers are useful in sorting out those males who tested with similar repeat counts for their markers and determining the probability of how close the common ancestor was.

The markers are usually inherited from father to son(s) unchanged, but a marker can still mutate to a shorter or longer repeat sequence. Every male with the same lineage will likely have a similar or same set of repeat counts, but the common ancestor may be a few or several to many generations back. The more STR markers tested, the better. If two males match on 25 markers, they could be off by several markers if they do a deeper 37-marker test. That means a difference of several repeat counts in a 37-marker test means the common ancestor was further back in time than might be suggested with the 25-marker match.

This table lists 76 named STR markers with Slow, Medium, and Fast mutation (S,M,F) indicators, the most useful are highlighted in red. Two testing companies were used, providing a combined total of 76 markers. Each testing company has its own database of test results. There are also free, public databases, such as ysearch.org and ybase.org, to find matches and compare results.

So far, no close matches have been found. The best matches have a genetic distance of 1 at 25 markers, and 7 at 67 markers. So, we are related through a common ancestor 24 or more generations back (about 600 years at 25 years per generation). However, as the databases grow, the potential for finding better matches increases. The Genetic Distance count is where an STR marker repeat count is down by one or more, or up by one or more. A NULL repeat count means there was an anomaly that prevented an accurate count so further specialized testing is required.
